Does Tone Match function in Axe/Power amp/Cab setup?

So, assuming Cab sims are disabled as is recommended in power amp/cab setup, will the TM block still function?

If so, are there any extra steps one has to follow to get it to sound/perform correctly?

My setup is Axe FX II/VHT 2|50|2/Mesa 2x12

Someone else asked this question in another post. I currently use a Mosvalve MV-962 Poweramp with two Marshall 4x12 cabs. I was so impressed with the Tone Matching results thru headphones that I might buy a pair of monitors.
