When to enable or disable the mic in the cab block?

Depends... Most current IRs have the mic(s) baked in, so you don't really need that.

More old school approach was to create the IRs with "neutral" mics and add the mic "flavor" later.

Ultimately it's just an EQ. Try it and use it if you like it.

On the Axe Fx III, FM9 and FM3 there are none anymore.
 
Thank you. That was the more in depth response I was hoping for. Unfortunately, I don't have a DAW and studio monitors to try out the differences. I have to use our mains for our P.A. (QSC K12), to try out the sounds. It is a bit loud for my apartment. I'm trying to keep the tweaking to a minimum at band practice. I appreciate your response.
 
I plugged in to my K12 yesterday. I tried both Enable and Disable. I didn't hear much difference, so I went ahead and disabled all mics on my presets.
