Any major re-tweaks going 6.0 to 6.01?

RTFRN for 6.01 :p

Just a small general adjustment to X-Former Match (easily tweaked back)
"The Transformer Match internal values have been reduced. This may be detectable as a slightly more open and less compressed tone. If you desire the slightly more compressed sound of Version 6.00, this can be obtained by increasing the Transformer Match to a value of 1.1."
and
Cliff: The Virtual Triode internal parameter values were tweaked slightly based on matching studies. In most amps this has increased the harmonic density. The aural effect is that the tones are a bit richer and smoother even though the frequency response is unchanged.

6.02 will be out "shortly"...

IMO, the amount of rework is really dependent on your preset creation style.

The more advanced params that you tweak, the more rework.

If you can wean yourself off of tweaking the advanced params, the firmware upgrades are less painful, and to my ears, the tones are actually noticably better. I attribute that to picking up all of Cliff's tweaks vs. my own tweaks.

Tweaks are unnecessary assuming you didn't have massive tweaks in the advanced section of the amp block... IMHO, no.
 
Just curious if I will have to redo everything or not really with this upgrade?

the only thing you need to do with 6.01 is turn up the volume and enjoy it...
and if you handle the volume side of things correctly...
the rest of the neighborhood can enjoy it too..
